The deduced amino acid sequence of Flac1 with 496 amino acids shows 58-64% homologies with other fungal laccases. The recombinant Flac1 (rFlac1) was heterologously expressed in Pichia pastoris, with sugars of approximately 4 kDa attached on the protein molecule, which has the calculated molecular mass of 53,532 Da. rFlac1 was shown to be a multi-copper oxidase from spectroscopies. The optimum pHs of rFlac1 for oxidations of 2,2\u0027-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), p-phenylenediamine, and o-aminophenol, were 5.0, 5.0, and 6.0-6.5, respectively, showing higher pH values than those from many other fungal laccases. The slightly acidic or neutral optimum pH that is not strongly dependent on substrates is a unique property of rFlac1. Effective O2 reduction was realized by the direct electron transfer of rFlac1 at a highly oriented pyrolytic graphite electrode modified with fine carbon particles (Ketjen Black) in O2-saturated solution. The pHs showing the maximum ΔE°\u0027 [= E°\u0027(enzyme) - E°\u0027(substrate)] coincided well with the optimum pHs shown by rFlac1 under steady-state conditions. The present electrochemical results of rFlac1 indicate that ΔE°\u0027 is one of the primary factors to determine the activity of multi-copper oxidases. © 2012 The Society for Biotechnology, Japan", "subitem_description_type": "Abstract"}]}, "item_4_publisher_17": {"attribute_name": "出版者", "attribute_value_mlt": [{"subitem_publisher": "Society for Biotechnology, Japan (日本生物工学会) / Elsevier"}]}, "item_4_relation_12": {"attribute_name": "DOI", "attribute_value_mlt": [{"subitem_relation_type": "isVersionOf", "subitem_relation_type_id": {"subitem_relation_type_id_text": "10.1016/j.jbiosc.2012.09.011", "subitem_relation_type_select": "DOI"}}]}, "item_4_source_id_11": {"attribute_name": "NCID", "attribute_value_mlt": [{"subitem_source_identifier": "AA11307678", "subitem_source_identifier_type": "NCID"}]}, "item_4_source_id_9": {"attribute_name": "ISSN", "attribute_value_mlt": [{"subitem_source_identifier": "1389-1723", "subitem_source_identifier_type": "ISSN"}]}, "item_4_version_type_25": {"attribute_name": "著者版フラグ", "attribute_value_mlt": [{"subitem_version_resource": "http://purl.org/coar/version/c_ab4af688f83e57aa", "subitem_version_type": "AM"}]}, "item_creator": {"attribute_name": "著者", "attribute_type": "creator", "attribute_value_mlt": [{"creatorNames": [{"creatorName": "Saito-Otsuka, Kaori"}], "nameIdentifiers": [{"nameIdentifier": "15780", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Kurose, Shinji"}], "nameIdentifiers": [{"nameIdentifier": "15781", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Tsujino, Yoshio"}], "nameIdentifiers": [{"nameIdentifier": "15782", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Osakai, Toshiyuki"}], "nameIdentifiers": [{"nameIdentifier": "15783", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Kataoka, Kunishige"}], "nameIdentifiers": [{"nameIdentifier": "15784", "nameIdentifierScheme": "WEKO"}]}, {"creatorNames": [{"creatorName": "Sakurai, Takeshi"}], "nameIdentifiers": [{"nameIdentifier": "15083", "nameIdentifierScheme": "WEKO"}, {"nameIdentifier": "90116038", "nameIdentifierScheme": "e-Rad", "nameIdentifierURI": "https://kaken.nii.ac.jp/ja/search/?qm=90116038"}, {"nameIdentifier": "90116038", "nameIdentifierScheme": "研究者番号", "nameIdentifierURI": "https://nrid.nii.ac.jp/nrid/1000090116038"}]}, {"creatorNames": [{"creatorName": "Tamiya, Eiichi"}], "nameIdentifiers": [{"nameIdentifier": "15785", "nameIdentifierScheme": "WEKO"}]}]}, "item_files": {"attribute_name": "ファイル情報", "attribute_type": "file", "attribute_value_mlt": [{"accessrole": "open_date", "date": [{"dateType": "Available", "dateValue": "2017-10-03"}], "displaytype": "detail", "download_preview_message": "", "file_order": 0, "filename": "SC-PR-SAKURAI-T-159.pdf", "filesize": [{"value": "1.0 MB"}], "format": "application/pdf", "future_date_message": "", "is_thumbnail": false, "licensetype": "license_free", "mimetype": "application/pdf", "size": 1000000.0, "url": {"label": "SC-PR-SAKURAI-T-159.pdf", "url": "https://kanazawa-u.repo.nii.ac.jp/record/10420/files/SC-PR-SAKURAI-T-159.pdf"}, "version_id": "671d0c09-a4fe-4ae3-a80f-8fab598eddde"}]}, "item_keyword": {"attribute_name": "キーワード", "attribute_value_mlt": [{"subitem_subject": "Direct electron transfer", "subitem_subject_scheme": "Other"}, {"subitem_subject": "Flammulina velutipes", "subitem_subject_scheme": "Other"}, {"subitem_subject": "Heterologous expression", "subitem_subject_scheme": "Other"}, {"subitem_subject": "Laccase", "subitem_subject_scheme": "Other"}, {"subitem_subject": "Optimum pH", "subitem_subject_scheme": "Other"}, {"subitem_subject": "Redox potential", "subitem_subject_scheme": "Other"}]}, "item_language": {"attribute_name": "言語", "attribute_value_mlt": [{"subitem_language": "eng"}]}, "item_resource_type": {"attribute_name": "資源タイプ", "attribute_value_mlt": [{"resourcetype": "journal article", "resourceuri": "http://purl.org/coar/resource_type/c_6501"}]}, "item_title": "Electrochemical characterization of a unique, \"neutral\" laccase from Flammulina velutipes", "item_titles": {"attribute_name": "タイトル", "attribute_value_mlt": [{"subitem_title": "Electrochemical characterization of a unique, \"neutral\" laccase from Flammulina velutipes"}]}, "item_type_id": "4", "owner": "3", "path": ["937"], "permalink_uri": "http://hdl.handle.net/2297/33410", "pubdate": {"attribute_name": "公開日", "attribute_value": "2017-10-03"}, "publish_date": "2017-10-03", "publish_status": "0", "recid": "10420", "relation": {}, "relation_version_is_last": true, "title": ["Electrochemical characterization of a unique, \"neutral\" laccase from Flammulina velutipes"], "weko_shared_id": -1}

内容記述 | The flac1 gene consisted of 1488 bases encodes a novel laccase (Flac1) from Flammulina velutipes. The deduced amino acid sequence of Flac1 with 496 amino acids shows 58-64% homologies with other fungal laccases. The recombinant Flac1 (rFlac1) was heterologously expressed in Pichia pastoris, with sugars of approximately 4 kDa attached on the protein molecule, which has the calculated molecular mass of 53,532 Da. rFlac1 was shown to be a multi-copper oxidase from spectroscopies. The optimum pHs of rFlac1 for oxidations of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), p-phenylenediamine, and o-aminophenol, were 5.0, 5.0, and 6.0-6.5, respectively, showing higher pH values than those from many other fungal laccases. The slightly acidic or neutral optimum pH that is not strongly dependent on substrates is a unique property of rFlac1. Effective O2 reduction was realized by the direct electron transfer of rFlac1 at a highly oriented pyrolytic graphite electrode modified with fine carbon particles (Ketjen Black) in O2-saturated solution. The pHs showing the maximum ΔE°' [= E°'(enzyme) - E°'(substrate)] coincided well with the optimum pHs shown by rFlac1 under steady-state conditions. The present electrochemical results of rFlac1 indicate that ΔE°' is one of the primary factors to determine the activity of multi-copper oxidases. © 2012 The Society for Biotechnology, Japan | |||||
