But the research on In Vitro germ cell induction which could be a fundamental solution to the problem has been just started.\nIn this study, I combined the Primordial Germ Cell-Like Cell (PGCLC) induction technique and the immunoprecipitation - mass spectrometry analysis to perform extensive analyses and tried to understand the mechanism which regulate germ cell development in cooperation with transcription factors. 内容記述 | 我が国を含めた先進国における出生率の低下は早急に対処すべき問題であるのに対し、根本的な解決策となりうる生体外における生殖細胞誘導の研究は、端緒に就いたばかりである。 本研究では、転写因子と協調して生殖細胞分化を制御する機構を解明するため、始原生殖細胞様細胞 (PGCLC)誘導技術と免疫沈降法―質量分析法を組み合わせた大規模で網羅的な解析を行った。さらに、CRISPR/Cas9技術を用いて候補因子の欠損および、転写因子の結合するゲノム領域の欠失を引き起こし、PGCLC誘導過程で機能検証実験を行った。 その結果、生殖細胞の発生過程に必須な転写因子間の相互作用や、エピゲノムの詳細が明らかとなった。 |
