末梢体内時計の生理的・臨床的意義の解明と制御法の開発 Evaluation of the pathophysiological signicance of peripheral clocks and the development of methods of controlling them 50382875 安藤, 仁 末梢体内時計の生理的意義と制御機構を解明するために、肥満2型糖尿病モデルであるob/obマウスの体内時計を解析した。ob/obマウスの体内時計障害は、(1)中枢時計には認められず、末梢組織特異的に認められ、(2)代謝異常が出現する以前の3週齢時より存在し、(3)レプチンの短期間投与により改善した。したがって、末梢時計障害は代謝異常の結果としては生じていないこと、末梢時計の制御にはレプチンシグナルが重要であることが明らかとなった。 To elucidate the physiological significance and controlling mechanism of peripheral clocks, the biological clocks in obese diabetic ob/ob mice were studied. The impairment of circadian clocks in ob/ob mice was (1) found in the peripheral tissues, but not in the suprachiasmatic nucleus (central clock), (2) detectable before the development of metabolic abnormalities, and (3) improved by short-term treatment with leptin.
