神経膠芽腫腫瘍幹細胞における遺伝子異常の時間的・空間的解析 Genetic analysis of glioblastoma cancer stem cell with special reference to chronological and spatial change 90262568 林, 裕 神経膠芽腫の発生・悪性化における時間的・空間的特質を解析するために腫瘍幹細胞における特定因子の解析を行った。幹細胞マーカーであるCD133を用い陽性細胞の分離、培養に成功した。それらを用い空間的特質である浸潤能に関与するintegrin familyの発現形式を検討した。その結果4種類のαsubunitの中でα3subunitが有意に高い発現を示し、腫瘍浸潤能への関与が示唆された。また、神経膠芽腫の時間的特質である悪性化の過程でアポトーシスや幹細胞調節に関係するFAS遺伝子とTP53遺伝子の変異の関与が疑われた。 To evaluate the specific factors in tumorigenesis of the glioblastoma, especially in chronological and spatial character, several genes expressions and mutations were analyzed. Among the integrins that could be related with invasion, the alpha 3 subunit had a significantly higher expression level in tumor stem cells. The alpha 3 subunit of integrin could play an important role in the tumor invasion. Furthermore, during a course of tumor progression, FAS and TP53 mutation were detected.
