Theobromine works as a phosphodiesterase (PDE) inhibitor to increase intracellular cyclic adenosine monophosphate (cAMP). cAMP activates the cAMP-response element-binding protein (CREB), which is involved in a large variety of brain processes, including the induction of the brain-derived neurotrophic factor (BDNF). BDNF supports cell survival and neuronal functions, including learning and memory. Thus, cAMP/CREB/BDNF pathways play an important role in learning and memory. Here, we investigated whether orally administered theobromine could act as a PDE inhibitor centrally and affect cAMP/CREB/BDNF pathways and learning behavior in mice. The mice were divided into two groups. The control group (CN) was fed a normal diet, whereas the theobromine group (TB) was fed a diet supplemented with 0.05% theobromine for 30 days. We measured the levels of theobromine, phosphorylated vasodilator-stimulated phosphoprotein (p-VASP), phosphorylated CREB (p-CREB), and BDNF in the brain. p-VASP was used as an index of cAMP increases. Moreover, we analyzed the performance of the mice on a three-lever motor learning task. Theobromine was detectable in the brains of TB mice. The brain levels of p-VASP, p-CREB, and BDNF were higher in the TB mice compared with those in the CN mice. In addition, the TB mice performed better on the three-lever task than the CN mice did. These results strongly suggested that orally administered theobromine acted as a PDE inhibitor in the brain, and it augmented the cAMP/CREB/BDNF pathways and motor learning in mice. © 2016 Elsevier Inc.","subitem_description_type":"Abstract"}]},"item_4_description_22":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"Embargo Period 12 months","subitem_description_type":"Other"}]},"item_4_publisher_17":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"Elsevier"}]},"item_4_relation_12":{"attribute_name":"DOI","attribute_value_mlt":[{"subitem_relation_type":"isVersionOf","subitem_relation_type_id":{"subitem_relation_type_id_text":"10.1016/j.jnutbio.2016.10.002","subitem_relation_type_select":"DOI"}}]},"item_4_relation_28":{"attribute_name":"関連URI","attribute_value_mlt":[{"subitem_relation_type_id":{"subitem_relation_type_id_text":"http://www.elsevier.com/locate/issn/09552863","subitem_relation_type_select":"URI"}}]},"item_4_rights_23":{"attribute_name":"権利","attribute_value_mlt":[{"subitem_rights":"Copyright © Elsevier (CC-BY NC ND)"}]},"item_4_source_id_11":{"attribute_name":"NCID","attribute_value_mlt":[{"subitem_source_identifier":"AA10748929","subitem_source_identifier_type":"NCID"}]},"item_4_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"0955-2863","subitem_source_identifier_type":"ISSN"}]},"item_4_version_type_25":{"attribute_name":"著者版フラグ","attribute_value_mlt":[{"subitem_version_resource":"http://purl.org/coar/version/c_ab4af688f83e57aa","subitem_version_type":"AM"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"Yoneda, Mitsugu"}],"nameIdentifiers":[{},{},{},{}]},{"creatorNames":[{"creatorName":"Sugimoto, Naotoshi"}],"nameIdentifiers":[{},{},{},{}]},{"creatorNames":[{"creatorName":"Katakura, Masanori"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Matsuzaki, Kentaro"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Tanigami, Hayate"}],"nameIdentifiers":[{}]},{"creatorNames":[{"creatorName":"Yachie, Akihiro"}],"nameIdentifiers":[{},{},{},{}]},{"creatorNames":[{"creatorName":"Ohno-Shosaku, Takako"}],"nameIdentifiers":[{},{},{}]},{"creatorNames":[{"creatorName":"Shido, Osamu"}],"nameIdentifiers":[{}]}]},"item_files":{"attribute_name":"ファイル情報","attribute_type":"file","attribute_value_mlt":[{"accessrole":"open_date","date":[{"dateType":"Available","dateValue":"2018-01-02"}],"displaytype":"detail","filename":"ME-PR-SUGIMOTO-N-110.pdf","filesize":[{"value":"410.6 kB"}],"format":"application/pdf","licensetype":"license_note","mimetype":"application/pdf","url":{"label":"ME-PR-SUGIMOTO-N-110.pdf","url":"https://kanazawa-u.repo.nii.ac.jp/record/14012/files/ME-PR-SUGIMOTO-N-110.pdf"},"version_id":"5ed4bb91-d89f-4d65-863e-8a86641846f8"}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"Theobromine up-regulates cerebral brain-derived neurotrophic factor and facilitates motor learning in mice","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"Theobromine up-regulates cerebral brain-derived neurotrophic factor and facilitates motor learning in mice"}]},"item_type_id":"4","owner":"3","path":["1134"],"pubdate":{"attribute_name":"公開日","attribute_value":"2017-12-05"},"publish_date":"2017-12-05","publish_status":"0","recid":"14012","relation_version_is_last":true,"title":["Theobromine up-regulates cerebral brain-derived neurotrophic factor and facilitates motor learning in mice"],"weko_creator_id":"3","weko_shared_id":-1},"updated":"2023-07-27T17:47:14.651351+00:00"}
