{"created":"2023-07-27T06:29:55.843136+00:00","id":14738,"links":{},"metadata":{"_buckets":{"deposit":"f4ed3ad9-bdfd-4268-bced-ca860408bda5"},"_deposit":{"created_by":3,"id":"14738","owners":[3],"pid":{"revision_id":0,"type":"depid","value":"14738"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00014738","sets":["1132:1133:1134"]},"author_link":["20219","20168"],"item_4_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2005-10-31","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"5","bibliographicPageEnd":"342","bibliographicPageStart":"333","bibliographicVolumeNumber":"28","bibliographic_titles":[{"bibliographic_title":"日本臨床免疫学会会誌 = Japanese journal of clinical immunology"}]}]},"item_4_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"B細胞の生存・分化・抗体産生に重要な役割を果たすBAFF (B cell activating factor belonging to the tumor necrosis factor family）はTNFファミリーに属する分子で単球，マクロファージ，樹状細胞の細胞膜上に発現され，可溶型として分泌される．BAFFの受容体にはBAFF-R (BAFF receptor), BCMA (B-cell maturation antigen）およびTACI (transmembrane activator and calcium-modulator and cyclophilin ligand interactor）の3種類が知られておりいずれもB細胞の広範な分化段階において発現がみられる．BAFFシグナルは主にBAFF-Rを介して伝えられ，TACIは抑制性のシグナルを伝達している．BAFFはB細胞上の受容体との結合により未熟B細胞の生存と分化，成熟B細胞の増殖，自己反応性B細胞の生存を制御する．BAFF過剰発現マウスでは全身性エリテマトーデス（systemic lupus erythematosus ; SLE）やSjögren症候群に類似した症状を呈する．さらにSLE自然発症モデルマウスや関節リウマチ（rheumatoid arthritis ; RA）モデルマウスであるコラーゲン誘導関節炎においてBAFFアンタゴニストの投与にて症状が改善することが明らかにされた．そしてSLEやRA，Sjögren症候群，全身性強皮症の患者において血清BAFF濃度の上昇が報告されている．BAFFは末梢性B細胞の分化・生存に影響することから，BAFF/BAFF受容体の異常が末梢性トレランスの破綻を来たし，リウマチ性疾患の発症に関与していると推測される．近年SLEやRAにおいてB細胞をターゲットした治療が脚光を浴びており， BAFFが有望な治療標的となることが期待されている．　B cell activating factor belonging to the tumor necrosis factor family (BAFF) is a tumor necrosis factor (TNF) superfamily member best known for its role in the survival and maturation of B cells. BAFF is a ligand for three TNF receptor superfamily members : B-cell maturation antigen (BCMA), transmembrane activator and calcium-modulator and cyclophilin ligand interactor (TACI), and BAFF receptor (BAFF-R). Among them, BAFF-R plays the central role in the BAFF system, whereas TACI plays the inhibitory role. BAFF/BAFF receptors appear to span nearly all stages of B-lineage differentiation, ranging from the development, selection, and homeostasis of naive primary B cells to the maintenance of long-lived bone marrow plasma cells. Furthermore, excessive BAFF rescues self-reactive B cells from anergy, which may play a crucial role in the induction and development of autoimmunity. Mice overexpressing BAFF exhibit increased B cell numbers in spleen and lymph node and autoimmune phenotype similar to patients with systemic lupus erythematosus (SLE) and Sjögrens syndrome. Furthermore, inhibition of BAFF by TACI-Ig and BAFFR-Ig has been successful in treating murine models of SLE and rheumatoid arthritis. In humans, previous reports have shown elevated serum BAFF levels in SLE, rheumatoid arthritis, Sjögrens syndrome, and systemic sclerosis patients.
