{"created":"2023-07-27T06:44:36.110546+00:00","id":34884,"links":{},"metadata":{"_buckets":{"deposit":"622c38db-1f32-46f4-862e-1ff4e031c104"},"_deposit":{"created_by":3,"id":"34884","owners":[3],"pid":{"revision_id":0,"type":"depid","value":"34884"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00034884","sets":["2812:2813:2837"]},"author_link":["24980","15561"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1997-03-01","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"3p.","bibliographicVolumeNumber":"1995-1996","bibliographic_titles":[{"bibliographic_title":"平成8(1996)年度 科学研究費補助金 基盤研究(B) 研究成果報告書"},{"bibliographic_title":"1996 Fiscal Year Final Research Report","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"我々は組織再構築に関与する新しいMMP遺伝子を検索する過程で、従来知られている可溶性型のMMPと異なった膜貫通構造を持つ新しいクラスのMMPが存在することを遺伝子クローニングによって示した。この膜型MMP (MT-MMP)はがん細胞の表面に発現しており、インビトロの実験によってゼラチナーゼAの潜在型酵素の活性化を引き起こすことが示された。悪性癌細胞の表面にも発現していることが示された。 MT-MMPの本来の役割を明らかにする目的で、マウスの発生過程でのMT-MMP発現をゼラチナーゼAの発現と活性化との関連で調べることにより、MT-MMPが発生過程での器官形成にもゼラチナーゼAの活性化因子として重要な役割を果たしていることを示そうとした。 その結果、MT-MMPはMMP-2, TIMP-2と共に個体の発生段階で器官形成に伴って発現し、成熟に伴って減少する。MT-MMPの発現はほとんどの臓器・器官で認められ、成長の停止に相関した発現の現象が見られた。脳及び心臓は出生までにほぼ器官の成熟が見られる臓器であり、ここでは発現の変化は顕著ではなかった。大腿骨、肋骨、肺、腎臓での発現が特に高かった。MMP-2も同様な成長に伴った変化が見られたが、腎臓での発現の急激な低下と肝臓と脳での発現が低いのが特徴であった。これに対してTIMP-2は成長に伴う発現変化が明らかではないことが特徴であり、肝臓での発現が低かった。胎児組織でのMT-MMPの発現を免疫染色およびin situ hybridization法で調べた。発現場所は骨形成部位、心臓、筋肉、血管の主として間葉系細胞であり、組織レベルでもMT-MMPとMMP-2, TIMP-2の共発現が認められた。","subitem_description_type":"Abstract"},{"subitem_description":"Matrix metalloproteinases (MMPs) are pivotal enzymes for tissue remodeling of multicellular organisms by degrading the components of extracellular matrix. Since MMPs are secreted as inactive zymogens (pro-MMPs), they have to be activated in the tissue to function. However little is known about physiological activators in vivo where active tissue remodeling is taking place. We recently identified a membrane-type MMP (MT-MMP) as an activator of pro-MMP-2 (pro-gelatinase A=72-kDa type IV pro-collagenase) that is expressed in invasive tumors and is responsible for degradation of type IV collagen in the basement membrane. In this study we examined whether MT-MMP plays a role in activating pro-MMP-2 during mouse embryogenesis. MT-MMP expression was increased together with that of MMP-2 and its inhibitor TIMP-2 in embryos according to the days after gestation and decreased with maturation after birth.MT-MMP transcripts and the product were localized in the cells of the occificating tissues where both MMP-2 and TIMP-2 were detected. Activated MMP-2 was detected in the lysate prepared from the microdissected tissue where expression of the three genes were observed.
