{"created":"2023-07-27T06:50:16.617736+00:00","id":42995,"links":{},"metadata":{"_buckets":{"deposit":"89eccda4-e745-4380-bef1-dc14e443e172"},"_deposit":{"created_by":18,"id":"42995","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"42995"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00042995","sets":["2812:2813:2831"]},"author_link":["69180","69181"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2003-05","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"5p.","bibliographicVolumeNumber":"2001-2002","bibliographic_titles":[{"bibliographic_title":"平成14(2002)年度 科学研究費補助金 基盤研究(C) 研究成果報告書"},{"bibliographic_title":"2002 Fiscal Year Final Research Report","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"ITS-PCR法とマイクロチップ電気泳動法による病原微生物の迅速同定に関する研究をCorynebacterium属、Bacillus属、Mycobacterium属、Clostridium属、真菌、Nocardia属を中心に行った。Corynebacterium属7菌種10株は泳動パターンから7種類に分けられた。C.diphtheriaeの4株は特徴的なパターンを呈し、非病原性Corynebacteriumと容易に区別された。Mycobacterium属10菌種24株では1〜5本のバンドがみとめられ、計13種類のパターンに分類された。M.tuberculosis、M.avium、M.intracellularの3菌種では2種類のパターンがみられたが、これらの菌種の鑑別はできなかった。その他のMycobacterium属10菌種のうち6菌種は菌種に特徴的なパターンを呈し、迅速同定に有用と思われた。Clostridium属5菌種は菌種ごとに異なるパターンを呈し、特にC.difficile36株は26種類に分類され、毒素(トキシンA、B)を産生する4株は4パターンに分けられた。真菌25菌種はMultiplex PCR法により、すべての菌株で2本のバンドがみとめられ、その泳動パターンから酵母菌種の同定が可能であった。しかし、Aspergillus属3菌種5株とNocardia属3菌種3株は属レベルの同定は可能であったが、種レベルの同定はできなかった。\n平成15年2月までに提出されたグラム染色陽性血液培養ボトル250検体のうち2菌種以上分離された5検体を除く245検体において、グラム染色所見を参考にすることにより属レベルまたは菌種レベルの同定が可能であった。今後、菌株を増やして、精度の高いデータベースを構築するとともに、迅速PCR法を用いて1時間以内に病原菌の同定を可能にしたいと考えている。","subitem_description_type":"Abstract"},{"subitem_description":"PCR amplification of the 16S-23S internal transcribed spacer (ITS) followed by microchip electerophoresis was evaluated for the usefulness in rapid identification of bacteria. Multiplex PCR amplification was used to test fungi, and Nocardia strains : the ITS 1 and 2 regions and the 5.8S rDNA region were amplified by using universal primers ITS1 and ITS4. The ITS2 region was simultaneously amplified by using universal primers ITS3 and ITS4.\nITS-PCR technique used in this study correctly identified clinically important bacterial strains, and Candida strains at species level within 2 h. Twenty-six different ITS-PCR patterns were demonstrated for 36 Clostridium difficile isolates on the basis of differences in the length of one or more amplicons. In addition, 202 Staphylococcal strains were divided into 40 ITS-PCR patterns. Therefore, the analysis of ITS-PCR patterns of C.difficile and staphylococci may be useful tool for epidemiological study of these strains.\nThe ITS-PCR method, prospectively applied to Gram staining-positive blood culture bottles, resulted in the correct detection and identification of bacterial strains and yeast strains.
