{"created":"2023-07-27T06:50:16.883411+00:00","id":43001,"links":{},"metadata":{"_buckets":{"deposit":"4f068ba7-6a03-440e-993d-c2a99acb09da"},"_deposit":{"created_by":18,"id":"43001","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"43001"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00043001","sets":["2812:2813:2838"]},"author_link":["20060","69186"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1996-03","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"11p.","bibliographicVolumeNumber":"1993-1995","bibliographic_titles":[{"bibliographic_title":"平成7(1995)年度 科学研究費補助金 一般研究(C) 研究成果報告書"},{"bibliographic_title":"1995 Fiscal Year Final Research Report","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"ヒトセミノーマ移植腫瘍におけるPLAP濃度はHeLa細胞移植腫瘍より高いにもかかわらずAnti-PLAP MAbの集積性は低く腫瘍イメージングは不可能であった。そこでこれまでにCEAなどにおいて報告されているIFN投与による細胞膜抗原の発現増強効果がヒトセミノーマ移植腫瘍のPLAPにおいて誘導されうるか否かを検討した。IFN投与により腫瘍組織内PLAP濃度は約50%増加した。抗体の腫瘍集積性は増強し抗体投与11日後において腫瘍イメージの改善をみた。しかしこの際の腫瘍/血流 比は1.0以下で抗体投与後早期から腫瘍イメージを捕えることは困難であり、INF投与量や投与方法に検討の余地を残した。\nついでSCIDマウス精巣にHeLa細胞をinjectionすることにより精巣腫瘍およびそのリンパ節転移モデルを作製し、Anti-PLAP MAbの血行性およびリンパ行性投与における腫瘍集積性について検討した。この精巣腫瘍転移モデルでは肝、肺にヒトベータグロビンのPCR産物として検出されるmicrometastasisの存在が示された。そこでMacro-and micrometastasisに対するI-131 labeled Anti-PLAP MAbを用いたRadioimmunotherapyによる抗腫瘍効果を検討した。抗体の腫瘍集積性は精巣腫瘍よりもリンパ節転移巣に高く、これは病巣の大きさおよび抗原量の相違によるものと考えられた。また抗体の腫瘍集積の絶対量は血行性投与の方が勝っていた。リンパ行性投与によるニンパ節に対する優位な抗体の集積は認められずこの転移モデルにおいては腫瘍局在診断および標的治療に際し、抗体の血行性投与の有用性が示唆された。精巣腫瘍およびリンパ節転移巣に対しては腫瘍重量においてそれぞれ57%、95%の抑制率を認めたが、組織学的に残存腫瘍が確認された。またこのモデルにおいては腫瘍細胞注入後1週目より肝、肺にPCRによって検出可能であるmicrometastaisが認められたことより、これらの病巣に対する抑制率を検討したところ、それぞれ97%、81%であった。以上の結果よりPI標識抗体を用いたRadioimmunotherapyの抗腫瘍効果が確認されたが、PCRでのみ検出されるmicrometastasisに対しても同療法の治療限界が示唆された。","subitem_description_type":"Abstract"},{"subitem_description":"We investigated significance of placental alkaline phosphase (PLAP) as a tumor marker for seminoma, and assessed a potential of PLAP as a targeting antigen for immunodetection and immunotherapy using anti-PLAP MAb. The monitoring of serum PLAP might be of value, as fluctuations in this marker provide information adout disease status and prognosis. Specific localization of anti-PLAP MAb to PLAP-producing xenografts was confirmed in a nude mouse model. In a SCID mouse model of testicular tumor and its lymph node metastases, localization of the MAb in lymph node metastases was more efficient than localization in testis tumors, suggesting the potential use of anti-PLAP MAb for radioimmunotherapy (RIT) on small metastatic foci of seminoma.
