{"created":"2023-07-27T06:50:22.538686+00:00","id":43129,"links":{},"metadata":{"_buckets":{"deposit":"363e12db-87e6-4227-9e2e-8f190997848d"},"_deposit":{"created_by":18,"id":"43129","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"43129"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00043129","sets":["2812:2813:2842"]},"author_link":["69348","69349"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1992-03","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"5p.","bibliographicVolumeNumber":"1990-1991","bibliographic_titles":[{"bibliographic_title":"平成3(1991)年度 科学研究費補助金 一般研究(C) 研究成果報告書"},{"bibliographic_title":"1991 Fiscal Year Final Research Report","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"細胞性因子によるアデノウイルス12型(Ad12)DNAの転写と複製の調節機構について以下に述べる研究成果が得られた。\n1.Ad12DNAの転写と複製を調節する細胞性因子の同定。\n(1)NFーIの同定。\nAd12E1A遺伝子の2つの転写のうちD転写を特異的に促進する因子をエ-ルリッヒ腹水癌細胞の核抽出液中に見い出し、精製を行った。精製標品は48,53,55kDaの3つのペプチドを含んでいた。この因子はAd12DNAの左端を1番としたとき22番〜48番の領域に結合したが、この配列はHeLa細胞のNFーIが認識する配列と一致した。さらにこの因子は、Ad2DNAの複製開始反応とHeLaNFーIと同程度に促進した。よってこの因子はマウスNFーIであると結論された。マウスNFーIはAd12DNAの左端付近に結合することにより、転写と複製両反応を調節すると考えられる。\n(2)ESFー1の同定。\nHeLa細胞の核抽出液中に存在するESFー1と名づけた因子が、Ad12E1A遺伝子のP転写を促進することが分った。この因子はTATAボックス直前の5'ーTGTCAー5'配列に結合する。\n2.Ad12E1A遺伝子転写の自己調節。\nDNAトランスフェクションにおいて、135mRNA由来のE1Aタンパクが自己遺伝子転写を約10倍促進することが分った。12SmRNA由来のタンパクは影響を与えなかった。E1Aタンパクの作用を解析するためにAd12E1Aタンパクの大量生産を、酵母と用いた系により行った。現在まで粗抽出液中にそれらしいペプチドを検出でき、抗体による確認を行っているところである。得られたE1AタンパクとNFーI、ESFー1との相互作用を調べる予定である。","subitem_description_type":"Abstract"},{"subitem_description":"Celluar factors that regulate transcription and replication of adenovirus type-12(Adl2)DNA were analyzed.\n1 Identification of cellular factors regulating transcription and replication of Adl2 DNA.\n(1) Identification of nuclear factor I (NF-I) that stimulates distal transcription of the AD12 E1A gene and replication of Ad DNA.\nA factor stimulating Adl2 EIA gene transcription in vitro was purified to near homogeneity from nuclear extracts of Ehrlich ascites tumor cells. A purified factor contained three peptides with molecular masses of 48, 53 and 55 kDa. This factor bound to a region located at the left end of Adl2 DNA where NF-I of, HeLa cells bound. Moreover, a purified factor stimulated replication initiation of Ad2 DNA in'vitro to the same extent as HeLa NF-I did. We thus concluded that a purified factor is mouse NFI. These results indicate that NF-I regulates both transcription and replication of Ad 1 2 DNA - by binding to the left end of the viral genome.\n(2) Identification of ESF-1 that stimulates proximal transcription of the Adl2 ElA gene.\nA factor stimulating proximal transcription of the Adl2 ElA gene was found in nuclear extracts of HeLa cells. This factor, designated ESF-1, recognizes a sequence 5'-TOTCA-3' located in a region adjacent to a TATA box. Purification of ESF-I is in progress.\n2 Autoregulation of Adl2 EIA gene.\nElA protein derived from 13S, but not 12S. MRNA stimulated transcription of its own gene in a DNA transcription assay. This indicates that Adl2 EIA gene transcription is under a positive autoregulation.
