{"created":"2023-07-27T06:51:43.517885+00:00","id":45255,"links":{},"metadata":{"_buckets":{"deposit":"c8d4cd48-e5f4-4cf4-aada-2e5af7739d85"},"_deposit":{"created_by":18,"id":"45255","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"45255"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00045255","sets":["2812:2813:2827"]},"author_link":["71055","78738"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2007-05","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"76p.","bibliographicVolumeNumber":"2005-2006","bibliographic_titles":[{"bibliographic_title":"平成18(2006)年度 科学研究費補助金 基盤研究(C) 研究成果報告書"},{"bibliographic_title":"2006 Fiscal Year Final Research Report","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"低周波交流磁界が生体、特に神経系にどのような影響を及ぼすかを調べるためモデル動物である線虫C.elegansを用いて磁界曝露による遺伝子発現レベルの変化と行動解析を行った。C.elegansに対して0.5Tの磁界を2時間暴露後、RNAを抽出し、前年度行ったマイクロアレイによる包括的なスクリーニングから発現に差があると見られる神経系遺伝子を抽出し、再現性の高いRT-PCRによる磁界曝露、非磁界曝露でのRNA発現量を比較した。\nマイクロアレイの結果、神経系関連遺伝子の多くはCa^<2+>に関連しており中でもカルモジュリンキナーゼIIをコードするunc-43遺伝子は磁界曝露により2.7倍の発現減少が認められ、RT-PCRの結果では2.35倍の減少を認めた。このことからCa^<2+>濃度を制御するカルモジュリン経路関連遺伝子(ckk-1(CaMKK), crh-1(CREB), cmk-1(CaMKIV), tax-6,cnb-1(Calcineurin)及びitr-1(IP3R))についてRT-PCRにより磁界曝露による遺伝子発現を検討した結果、unc-43,itr-1,cnb-1及びtax-6遺伝子は優位にRNAの発現が減少(P<0.01)したが、その他のckk-1,cmk-1及びcrh-1も減少傾向にあった。\n磁界曝露による行動解析には、線虫C.elegansの化学物質に対する誘引・忌避行動を指標とした解析により影響を評価した。誘引物質であるDiacetyl、忌避物質であるCuSO_4を培地上の一端に置き、それぞれの物質への到達率をカウントしたところ磁界曝露によりDiacetylへの到達率が減少し、またCuSO_4に対する突破率が増加した。\nこのことから磁界はある特定の神経細胞に働き、Ca^<2+>濃度の変化を惹起させ遺伝子発現レベルを変化させることで行動に異常をもたらした可能性が示唆された。","subitem_description_type":"Abstract"},{"subitem_description":"The biological effect of extremely low frequency magnetic fields (ELFMFS) remains controversial. Some epidemiological studies have indicated positive correlations between magnetic fields and various types of cancer or other disease, including neurological disorders. To clarify the biological effects of ELFMFs, we performed the screening of ELFMFS-responding genes by using microarray technique in nematode C. elegans, as a model organism, and carried out the RT-PCR analysis not only the genes with significant difference identified by microarray, but also the associative and relative genes with possibility of ELFMFs-responding genes. Moreover, we investigated chemotactic response to either diacetyl or CuSO_4 after ELFMFs exposing.\nunc-43, one of the identified genes, is known to function as calmodulin-dependent protein kinase II (CaMKII), that mediates Ca^<2+> signal transmission in the nervous system of C. elegans. We found that unc-43 mRNA was down-regulated (2.4-fold) under ELFMFs exposure by RT-PCR.
