| 内容記述 | 骨肉腫の培養細胞に、後期糖化産物受容体（Receptor for advanced glycation end-products: RAGE）を強制発現させると幹細胞マーカーであるCD133、CD24の発現が亢進し、腫瘍塊形成能が高く、また抗がん剤に対する薬剤耐性能も高くなることが示された。手術切除標本におけるCD133の発現は、有意な予後不良因子であった。さらに網羅的遺伝子解析により、RAGE遺伝子導入により細胞周期、細胞分裂に関わる分子が発現誘導されることが判明した。今回の研究成果から、RAGEが骨肉腫においてがん幹細胞マーカーの発現に関与し、がん幹細胞の性質を生み出す可能性が示唆された。 | |||||

| 内容記述 | Induced expression of RAGE (receptor for advanced glycation end-products) resulted in the co-expression of CD133 and CD24, stem-cell markers, in human osteosarcoma cell lines. Moreover, the RAGE-overexpressing cells showed higher spheroid-forming ability and a resistance to treatment of cisplatin, an anti-cancer drug, in vitro.Upregulated gene expression of cell-cycle and cell-division drivers was observed in microarray analyses . We also found that the expression of CD133 was significantly correlated with poor prognosis in patients with osteosarcoma using immunohistochemical analyses. Our results suggest that the RAGE could elicit the expression of cancer stem cell markers and may promote the cancer stem-like cell characteristics in osteosarcoma. | |||||
