Lymphoproliferative Syndrome : ALPS)において増加するDN T細胞ではCD95/CD95L細胞死誘導欠損により調節性T細胞(Treg)機能が低下し、自己免疫病態が拡大する。DNT細胞のTreg細胞としての特徴を正常およびALPS患者において比較解析した。ALPSのDNT細胞の細胞表面発現はCD7、CD62L、CD45RO発現が低下、CD4+Treg細胞と同様にIL-7R陰性で、CD57, CD38, CD95, HLA-DR, CD45RO陽性であった。一方、CD4+Treg細胞の特異的転写因子FoxP3蛋白発現は認められなかった。DNT細胞の培養後細胞死比率はALPS83%、健常人30%と有意にALPSにおいて有意に増加していた。DNT細胞のTCRレパートワ分布解析ではALPS14症例中5例(35.7%)、正常対照40例中3例(75%)にTCRVβの1種類以上のあきらかな増加を認めた。CDR3spectrum解析ではALPS11例(78.5%)、健常人全例で3種類以上のVβにoligo-clonal patternを認めた。また、ALPSの症状を有するがCD95誘導性細胞死が正常であったALPS III型と考えられていた症例4例のうち1例にモザイク型CD95変異が存在することを証明した。変異比率はDNT細胞では100%、他のリンパ球分画でも1-10%存在し、繊維芽細胞や口腔粘膜上皮細胞にも1%存在し、モザイク型の変異を初めて見出した。","subitem_description_type":"Abstract"}]},"item_9_description_22":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"研究課題/領域番号:19591243,
