{"created":"2023-07-27T06:56:30.528878+00:00","id":53340,"links":{},"metadata":{"_buckets":{"deposit":"922632d9-484f-4c03-8c32-bd3fb46ff791"},"_deposit":{"created_by":18,"id":"53340","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"53340"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00053340","sets":["2812:2813:2821"]},"author_link":["157"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2013-05-20","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"6p.","bibliographicVolumeNumber":"2010 – 2012","bibliographic_titles":[{"bibliographic_title":"平成24(2012)年度 科学研究費補助金 基盤研究(B) 研究成果報告書"},{"bibliographic_title":"2012 Fiscal Year Final Research Report","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{},{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"従来のJakobssonらの提唱した浸潤様式分類の4型を4C型と4D型に細分類したわれわれの浸潤様式分類は、口腔扁平上皮癌の臨床での重要な予後因子となっている。本研究では in vitro および in vivo の両方の浸潤モデルを用いた浸潤増殖像の検討、および in vivo では浸潤モデルにおける浸潤像とリンパ節転移との関係についてそれぞれ検討した。また、口腔癌の臨床組織を用いた免疫組織化学的研究も行った。免疫染色の結果では浸潤様式が高度になるにつれて、癌胞巣周囲の基底膜の4型コラーゲンの連続性の消失と細胞間接着分子のE-cadherinの消失を認めた。また高度浸潤癌では各種のMMPの発現亢進と低酸素で発現するHIF-1αの発現の更新を認めた。In vitroの浸潤モデル実験には3種類の口腔扁平上皮癌細胞株OCS-20細胞 (浸潤様式3型), OSC-19細胞 (浸潤様式4C型), HOC-313細胞 (浸潤様式4D型)を用いた。その結果、コラーゲンゲルを用いた in vitro 浸潤モデルにおいて各癌細胞の浸潤能に応じた浸潤像を再現することに成功し、浸潤様式が高度になるにつれゲル中へのび漫性の浸潤を認めた。また、浸潤様式4D型のHOC-313細胞ではコラーゲンゲル中に線維芽細胞を入れない状態でも、同様にび漫性の浸潤を認めた。これらの細胞の運動能を検討した結果、浸潤能の高い細胞で浸潤時にAMFの高い発現を観察した。In vivoの浸潤実験では、マウスの舌に正所性移植するとOSC-20細胞と OSC-19細胞では、臨床で観察される浸潤像を再現することが可能であった。また、OSC-19細胞ではリンパ節転移も再現できた。HOC-313細胞では移植にmatrigelを使うことで一時的には移植が可能であった。さらに、OLC-01細胞を用いることで浸潤様式4D型の浸潤像を再現できることが判明した。","subitem_description_type":"Abstract"},{"subitem_description":"We have been studying the mode of cancer invasion as the important prognostic factor in patients with oral squamous cell cancer. Grade 4 of mode of invasion proposed by Jakobsson was subclassified into grades 4C (Cord-like type) and 4D (Diffuse type). The purpose of this study is to immunohistochemically and experimentally elucidate this modified grading system for the mode of invasion. Materials and methods used are human cancer materials and in vitro and in vivo invasion model by using 3 cell lines; OCS-20 (Gr.3), OSC-19 (Gr. 4C) and HOC-313 (Gr. 4D). In immunostained human cancer materials, the higher the grade of the mode of invasion was, the less continuous the type 4 collagen was, and the less E-cadherin was. Majority of diffuse invasive cancers (Gr. 4C and 4D) expressed MMPs and HIF-1. In cell invasion assay, HOC-313 showed the greatestmotility (AMF). Cell lines cultured on 3T3 fibroblast-embedded collagen gels showed similar invasion pattern to their origin. Among them, only HOC-313 showed invasion evenif fibroblast was not embedded. OSC-20 and OSC-19 implanted orthotopically in the tongue of nude mice grew and metastasized to lymph nodes. Their histologic appearance was similar to that of the origin. HOC-313 grew temporarily for several months by support of matrigel.
