Osamu"}],"nameIdentifiers":[{"nameIdentifier":"49997","nameIdentifierScheme":"WEKO"},{"nameIdentifier":"60193025","nameIdentifierScheme":"e-Rad","nameIdentifierURI":"https://kaken.nii.ac.jp/ja/search/?qm=60193025"}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"現在までの研究の結果から、肝細胞増殖因子(HGF)が種々の器官の初期発生において組織の分化に関わっていることがわかってきた。本研究においては、間葉系組織が軟骨、骨組織に分化するとともに、歯や舌など様々な組織発生を導き出す下顎の発生において、HGFの作用と生理的意義を解明することができた。すなわち、昨年度の研究において培地に加えたHGFが骨やメッケル軟骨の発生を促進すること、またこれらの効果が特異的受容体であるc-metにより制御されていることに加え、HGFのアンチセンスオリゴヌクレオチドを培養液に加えて本来組織に存在する内因性のHGF合成をブロックすると、メッケル軟骨の軟骨小腔が拡大して内部の軟骨細胞にアルカリ性フォルファターゼ活性が発現することがわかった。この現象は本来、軟骨内骨化によって軟骨が変性する際にみられるものである。またこのような軟骨の軟骨膜にも強いアルカリ性フォスファクターゼ活性が現れ、軟骨性の骨化に似た所見が認められた。このメッケル軟骨の骨化現象も子宮内の発生やコントロール群の培養下顎では通常この時期や場所ではおこらない。しかしアンチセンスオリゴとともにHGFを同時に添加するとこれらの現象は認められなかった。HGFのアンチセンスオリゴを骨化する軟骨である前肢の長骨原基の器官培養系に投与しても同様の変化を観察できた。従って組織内のHGFの濃度が減少すると、軟骨は変性して骨化を起こすことが示唆された。\n以上の結果から、軟骨の発生においてはHGFはその維持や増殖に働くとともに、骨化を抑制していることが強く示唆された。","subitem_description_type":"Abstract"}]},"item_9_description_22":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"研究課題/領域番号:09771497,
