cinnamoneum由来分泌性タンパク質Phospholipase D(PLD)の遺伝子および、その上流プロモーター領域と下流ターミネター領域を導入pUc702-promoter-PLDを構築し、Streptomyces lividansに組換えを施すことで、野生株に対し、約20倍のPLDの分泌生産を示した^<[1]>。本研究では、効率的なタンパク質生産やタンパク質の表層提示発現を目的として、本遺伝子組換え発現系の分泌生産に関与している分泌シグナル領域に様々な変異を導入しその発現挙動について解析を行った。また、放線菌由来の膜タンパク質をアンカーとしてPLDと融合発現させ放線菌におけるタンパク質の表層発現系の構築、Phospholipase A2(PLA2)などの有用タンパク質とPLDとの融合タンパク質発現系を構築した。その結果、以下の点について明らかとなった。\n1)放線菌ではGly-Xxx-Glyもシグナルペプチダーゼが認識してシグナルは切断される。\n2)PLD由来シグナルペプチドは疎水性が強くなることでタンパク質の生産性が向上することが明らかとなった。\n3)放線菌におけるPLD-PLA2融合タンパク質発現系を構築できた。また、codon
