capture microdissection法を用いて肝細胞や胆管細胞を切り出し、RNAを抽出し、リアルタイム(Taq-man)PCR法を用いて、Total RNA量の測定やHCV RNA量の定量などを検討してきた。前年度は、C型肝炎肝組織において、炎症の程度や小葉内の領域が異なっていても、HCV RNA量は特に偏在しないことを明らかにした。しかしRNAの質に関しての評価に関しては十分ではなかった。\nそこで今回は、肝凍結組織から回収したRNAの正確な量および質の評価のために、Agilent bioanalyzer nano-kitおよびpico-kitでの検討を追加した。本検討ではごく微量のRNA溶液(1μl)からそのRNA濃度とRNAの質を表すrRNA ratio (28S/18S)を、nanoおよびpico単位で測定可能である。約1.5cm×1.5cmの大きさで、10μmの厚さで切り出した肝組織を用いた検討では、300〜700ng/μlのRNAが10μl回収され、またrRNA ratio (28S/18S)は1.5〜2.0であり、良好な状態でRNAが回収されていることが確認された。さらに、Laser capture microdissection法にて肝小葉の半分ほどの大きさを切り出したものについても同様の検討を行ったところ、300〜400Pg/μlのRNAが10μl回収されたことが確認された。\n今回の検討により、凍結肝組織からLaser capture microdissection法を用いて切り出した細胞から抽出したRNAは、良質でかつ十分量であることが確認されたことから、引き続きRNA
