{"created":"2023-07-27T06:58:43.539223+00:00","id":57686,"links":{},"metadata":{"_buckets":{"deposit":"68f3d85f-3b46-421b-b225-f192c72f1616"},"_deposit":{"created_by":18,"id":"57686","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"57686"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00057686","sets":["2812:2813:2833"]},"author_link":["344"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2002-03-25","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"2p.","bibliographicVolumeNumber":"1999 – 2000","bibliographic_titles":[{"bibliographic_title":"平成12(2000)年度 科学研究費補助金 基盤研究(C) 研究成果報告書概要"},{"bibliographic_title":"2000 Fiscal Year Final Research Summary","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"グリア細胞が産生する神経成長因子(NGF)は脳神経細胞の生存を支え、神経突起の成長を引き起こす。遺伝子産物であるNGFの臨床応用を検討するにあたっては、生体内のNGF合成を促進する方法がNGFの直接投与よりも安全性を含めより現実的と考えられている。そこで本研究に置いては、担子菌・子嚢菌類からのNGF産生促進物質(プロモーター)の探索を行った。ヒトアストロサイトーマ(1321N1)からのNGF産生活性は、mRNAの発現量とPC-12細胞の神経突起成長によって測定した。本研究に先立って見出したscabronine Aは担子菌ケロウジ(Sarcodon scabrosus)から単離したことから、本年度は他の担子菌・子嚢菌類とともにケロウジの成分探索を進めた。各種クロマトグラフィー及び高速液体クロマトグラフィーによって分画・精製を進めた結果、新たにscabronine G,H及びIと名付けたジテルペノイドを単離し、2次元核磁気共鳴スペクトルおよび円二色性曲線の解析によって立体構造を決定した。Scabronine Gは1321N1細胞におけるNGF mRNA発現量を増加させ、scabronine Gとともに培養した1321N1細胞の培地を加えて培養したPC-12細胞は形態を変えるとともに、神経突起を引き起こした。Scabronine GはAとともに連続した5,6,7員環とカルボキシル基を持つことを特徴としており、それらの構造的特徴が活性発現に寄与しているのではないかと考えられる。2000年度に新たに単離したscabronine H及びIは同様の骨格を持つジテルペノイドである。強力なNGFプロモーション作用を持つscabronine AはC-15位についてのエピマーの混合物であったが、その11-エピマーであるscabronine HはC-15位に関してはS体のみが得られたことが興味深い。現在、scabronine H及びIのNGF産生活性については明確な実験結果が得られておらず、今後の精査が望まれる。","subitem_description_type":"Abstract"},{"subitem_description":"Novel cyathane-type diterpenoids, scabronine A-F, were isolated from the fruit body of the mushroom, Sarcodon scabrosus (Fr.) Karst. The stereostructures of those were elucidated on the basis of the spectroscopic analysis. Among these compounds, scabronines A, B, C and E exhibit inductive activity of the verve growth factor synthesis. Further investigation yielded three more congeners, scabronine G, H and I.\nWhen rat pheochromocytoma cells (PC-12) were cultivated with the conditioned medium of human astrocytoma cells (1321Nl) incubated with the new diterpenoids, scabronines A and G, isolated from Sarcodon scabrosus, they changed their morphology and there was neurite outgrowth. The scabronines increased the expression of nIRNA for nerve growth factor (NGF), and the secretion of NGF from 1321Nl cells in a concentration-dependent manner.
