vitroにて好中球、好塩基球、T細胞に対し強い走化活性を示すとともに、in vivoにて好中球依存性急性炎症における組織損傷に対して重要な役割を担っていることがわかっている。本研究において、IL-8をマウスを用いて特定のある臓器に特異的に過剰に発現させた場合、それが炎症性の疾患モデルとしての病態を呈することを想定し、tissue-specific(臓器特異的)、特に肝特異的なIL-8の発現マウス(IL-8 Tg)の作製とその解析を行った。\n導入遺伝子はマウス肝臓に特異的にサイトカインを発現させるために血清アミロイドP遺伝子(serum amyloid P component gene)のプロモーターを利用し(熊本大学、山村研より供与)、この下流にヒトIL-8cDNAを挿入し、これをマウス受精卵前核にマイクロマニピュレーターを用いて導入した。IL-8 TgはNo.53とNo.56の2系統を樹立した。ともにヒトIL-8をプローブにしたsouthern blotting(EcoRI処理)にて、約250bp付近にてシングルバンドを認め、northern blottingにて、肝臓特異的にヒトIL-8のmRNAの発現が認められた。\nまた、血清中のヒトIL-8量は4.0-10ng/mlに達し、末梢血白血球数はコントロールマウスと比較して有意の上昇を認めた(p<0.05)。好中球の割合もコントロールに比較して増加していた。末梢血白血球上の表面抗原(B220,CD3,Mac-1,Gr-1,ER,ERHi)はIL-8 Tgとコントロールにおいて明らかな差は見られなかった。しかし、末梢血好中球表面上のL-selectinはIL-8 Tgにおいてdownregulationが認められた。さらに、好中球表面上のmouse IL-8 receptorの発現はIL-8 Tgとコントロールにおいて差は認めなかった。病理組織学的にはIL-8
