{"created":"2023-07-27T07:00:57.923664+00:00","id":61082,"links":{},"metadata":{"_buckets":{"deposit":"6e636c64-ff70-4cc6-9a49-aca8a86682c9"},"_deposit":{"created_by":18,"id":"61082","owners":[18],"pid":{"revision_id":0,"type":"depid","value":"61082"},"status":"published"},"_oai":{"id":"oai:kanazawa-u.repo.nii.ac.jp:00061082","sets":["2812:2813:2842"]},"author_link":["26211","24592"],"item_9_biblio_info_8":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"1993-03-15","bibliographicIssueDateType":"Issued"},"bibliographicPageStart":"2p.","bibliographicVolumeNumber":"1990 – 1991","bibliographic_titles":[{"bibliographic_title":"平成3(1991)年度 科学研究費補助金 一般研究(C) 研究成果報告書概要"},{"bibliographic_title":"1991 Fiscal Year Final Research Report Summary","bibliographic_titleLang":"en"}]}]},"item_9_creator_33":{"attribute_name":"著者別表示","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{}],"nameIdentifiers":[{},{}]}]},"item_9_description_21":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"ヒト膀胱癌由来培養細胞株KKー47細胞よりHAT selectoonにて得られた細胞株は細胞感染が認められ,野性型細胞を用いて本研究を逐行した。光感受性物質はHpDI(Queen Elizabeths Hospital,Australia)を,培養液はEagle MEM(日水製薬)に10%中胎児血清(Microbological Association,USA)を加えたものを,照射光源はアルゴン色素レ-ザ-光(ADL,630nm,SpectraーPhysics)およびUV(UVA;300ー430nm,UVB;280ー320nm,東芝医療用機器株式会社,MーDMRー1)を用いた。HpDI処理はすべて暗室にて行った.HpDIは処理3時間で細胞毒性が認められない40μg/mlを用い,HpDI処理後ADL,lmW/cm^2,2分間照射(0.12J/cm^2)での細胞生残率は22%であり,以下この条件を用いた。UVA照射(5mW/cm^2)では6J/cm^2の照射量にて,UVB照射10.1mW/cm^2)では0.6mJ/cm^2にてそれぞれ約20%の細胞生残率であり,以下この照射量を用いた。突然変異細胞の選択薬として6ーthroguanine(6ーTG,Sigma)を用いた。10^6個/シャ-レの細胞にHpDIを接触させ,ADL照射した群,UVAまたはUVBのみを照射した群に分け,10^6個/シャ-レに分注し培養を続けた。この継代培養開始時を突然発現時間の0日目とした。継代培養0,2,4,6,8,9日目に細胞を取り出し,10^5個/シャ-レに分注し,5μg/mlとなるよう6ーTGを加え14日間培養した。同時にコロニ-形成法にて細胞生残率を求めた。UVB照射群における突然変異発現は4日目より増加し,6日目より9日目までほぼ一定であった。HpDI+ADL照射群およびUVA単独照射群の突然変異発現は9日目まで10^5個生細胞当たり8.0±52個とほぼ一定であり,無処理の対照群と変化なかった。HpDIを用いた光力学的作用には明らかなmutagenie activityは認められなかった。臨床的には,40例の難治性膀胱癌に対して光力学的治療を施行したがmutationを示唆する所見は認められなかった。","subitem_description_type":"Abstract"},{"subitem_description":"Cell culture studies have been performed to compare the mutagenic potential for hematoporphyrin derivative photoradiation and UV radiation. The mutation frequency in human bladder cancer cells (KK-47) at the hypoxanthine-guanine phosphoribosyltransferase locus was measured using resistance to 6-thioguanine. Treatment with UVB (wavelength ; 280-320 nm) was effective in producing mutants resistant to 6-thioguanine, but treatment with neither UVA (wavelength ; 300-430 nm) nor hematoporphirin derivative photoradiation (at comparable toxicity levels) using argon-dye laser (wavelength ; 630 nm) did not induce any mutagenic activity above background levels. Additional studies regarding hernatoporphyrin derivative UV radiation are needed.
