metalloproteinase 3(MMPー3=ストロムライシン)の関節組織内局在とMMPー3によるプロテオグリカンとIX型コラ-ゲンの分解について検討し、以下の新たな知見を得た。\n1.MMPー3はOA試料の83%(53/64例)で免疫組織学的に関節軟骨細胞に証明され、その局在は軟骨プロテオグリカンの消失と相関した。滑膜炎の合併した症例では、滑膜表層細胞もMMPー3を分泌していた。滑膜より精製されたMMPー3は、軟骨組織中のプロテオグリカンを分解することが光顕的及び電顕的に観察された。\n2.MMPー3はニワトリ硝子体由来のIX型コラ-ゲンを分解するのみならず、軟骨細胞より単離されたIX型コラ-ゲンのNCー4ドメインを分解した。\nこれらの成果に関して現在論文執筆中である。MMPー3の生化学的性質,活性化機序,RA関節組織における局在について既に報告してきた。それらに加えて、今回の新たな知見はRAとOAの関節軟骨破壊における本酵素の重要性を示している。\nIX型コラ-ゲンのRAとOA関節軟骨における局在に関しては,特異的ポリクロ-ナル抗体を作製して免疫染色を行ったが、はっきりしたデ-タは得られなかった。現在、プロテア-ゼ処理などの条件を検討中である。また、in situハイブリダイゼ-ションによるRAとOA関節軟骨におけるIX型コラ-ゲン合成能の検討は、インサ-トからIX型,X型,II型コラ-ゲンのcDNAを単離した状態であり、今後検討する予定である。","subitem_description_type":"Abstract"}]},"item_9_description_22":{"attribute_name":"内容記述","attribute_value_mlt":[{"subitem_description":"研究課題/領域番号:02670134,
