| 内容記述 | 本研究では, ドーピング薬に指定されているクレンブテロールを雄マウスに投与し, 骨格筋の乳酸の代謝特性と収縮特性に及ぼす影響について解析した.得られた結果は次の通りである.<BR>(1) 成長に及ぼす影響として, 投与群に体重の有意な増加が認められた.<BR>(2) 投与群の筋湿重量と体重比は速筋のEDLで差は認められず, 遅筋のSOLでは有意な増加が認められた.<BR>(3) 乳酸の代謝特性に関しては, 投与群の遅筋のSOLにおいてLDH比活性の上昇傾向とLDH総活性値の有意な上昇がみられ, Lactate shuttleにおける乳酸生成の促進が示された.また, 投与群の遅筋のSOLにおいてはLDHのMTypeの増加も認められ, 明らかな乳酸生成の促進を裏付けるものであった.速筋のEDLではいずれも変化は認められなかった.<BR>(4) 収縮特性に関しては投与群の遅筋のSOLにおいてMyosin-HCのType IIの割合が増加し, 筋全体の速筋化が認められた.しかし, 投与群の速筋のEDLにおいては, Myosin-HCでは影響は認められなかった.<BR>クレンブテロール投与により, 速筋のEDLよりも遅筋のSOLにおいて著明な速筋化がおこった.この結果は, β2レセプターにクレンブテロールが結合し, A-kinase系の反応が促進されたことで, 乳酸生成の促進と筋肥大をともなうタンパク合成の促進またはタンパク分解抑制, あるいはそれらの相互作用の結果, 筋線維タイプ移行が生じたためと推察される.<BR>クレンブテロールは主に, β2レセプターが遅筋により多く分布することから, 特に遅筋により強く作用すると推察された.しかし, 遅筋における筋肥大の直接的な作用機序, 核内の転写調節因子, MyoDやmyogeninの関与についてはまだわかっておらず, 今後の検討課題である. A study was conducted to investigate the effects of the beta-2 receptor agonist, clenbuterol, on the slow-twitch soleus (SOL) and the fast-twitch extensor digitorum longus (EDL) muscles. Twelve male 5-wk-old mice (ddY ) were divided into two groups ; control ( CANT, n=6 ) and clenbuterol-treated (CLEB, n=6) . Clenbuterol was given in the drinking water (0.02 mg/mL) for 5 weeks. Post-treatment body weights were approximately 10% greater in the CLEB group compared to CANT (P<0.05) . In the SQL muscles of the CLEB group the wet weight and the ratio of muscle weight to body weight was significantly higher than the CANT (P<0.01), but not in the EDL. In the SOL muscles of the CLEB group the muscle type LDH isozyme distribution and enzymatic activity was significantly increased (P<0.01) . However, there was no metabolic change in the EDL muscles. Polyacrylamide gel electrophoresis (SDS-PAGE) of myosin heavy chain (Myosin-HC) indicated a clenbuterol-induced decrease (P<0.05) in the relative percentage of type I Myosin-HC with a concomitant increase (P<0.05) in Type II Myosin-HC in the SQL of the CLEB group. The Myosin-HC composition in the EDL was not altered. These findings suggest that clenbuterol may have induced selectively changes of muscle hypertrophy, the predominant anerobic glycolysis, and the transition of Myosin-HC in the SOL. The differences in the two muscles can be explained by the higher number of beta-2 receptors in slow-twitch muscles. | |||||||||
