| 内容記述 | 病態形成におけるフラクタルカインの役割は、疾患ごとに炎症を惹起または抑制のいずれにも調節し、大きく異なる。本研究課題では、肥満による炎症の誘導と維持におけるフラクタルカインとその受容体CX3CR1の役割について検討を行った。受容体CX3CR1を欠損したマウスは、対照群に比し、高脂肪食摂取によるインスリン抵抗性・高血糖・脂肪肝の増悪を認めた。フラクタルカイン-CX3CR1系は、肥満によるインスリン抵抗性の形成に重要であることが示唆される。現在、炎症細胞との関連性に着目し、フラクタルカイン-CX3CR1の慢性炎症、インスリン抵抗性に与える影響とそのメカニズムについて解析を進めている。 | |||||

| 内容記述 | The physiological and pathophysiological role of fractalkine and its receptor CX3CR1 in diseases are complex and not fully understood. In particular, it is not known how fractalkine-CX3CR1 can regulate obesity-associated chronic inflammation and metabolic diseases. Here, we investigated the role of fractalkine and CX3R1 system in obesity-induced inflammation and insulin resistance. We found that CX3CR1 deficient mice showed that insulin resistance, glucose intolerance, and hepatic steatosis in response to high fat (HF) feeding. These data suggested that fractalkine-CX3CR1 signal play a crucial role in development of insulin resistance. Further studies to clarify the mechanism by which fractalkine-CX3CR1 regulate inflammation and insulin resistance are going on. | |||||
